Overview

Anti-BCMA 4-1BBz CAR-iNKT is an experimental, allogeneic cell therapy consisting of invariant natural killer T (iNKT) cells engineered to express a second-generation chimeric antigen receptor (CAR) targeting B-cell maturation antigen (BCMA). The construct utilizes a 4-1BB (CD137) co-stimulatory domain and a CD3ζ signaling domain. iNKT cells are uniquely suited for off-the-shelf use because their T-cell receptors (TCRs) are CD1d-restricted and do not cause graft-versus-host disease (GvHD), eliminating the need for TCR gene editing. In comparative academic studies for multiple myeloma, this specific 4-1BBz construct was evaluated alongside other designs (such as CD28z), with research suggesting that while functional, it may exhibit lower avidity and anti-tumor efficacy compared to CD28z-based CAR-iNKT cells.
