Overview

Anti-BCMA AlloCAR T refers to a suite of allogeneic chimeric antigen receptor (CAR) T-cell therapies developed by Allogene Therapeutics that target B-cell maturation antigen (BCMA). Unlike autologous CAR T therapies, which use a patient's own cells, these 'off-the-shelf' products are derived from healthy donor T-cells. The platform utilizes TALEN gene-editing technology to disrupt the T-cell receptor (TCR) alpha constant (TRAC) gene to reduce the risk of graft-versus-host disease (GvHD) and the CD52 gene to allow for selective lymphodepletion. Specific candidates include ALLO-715 and ALLO-605, the latter being a 'TurboCAR' variant designed to enhance T-cell activation and persistence. These therapies are primarily being developed for the treatment of relapsed or refractory multiple myeloma.
