Overview

Anti-BCMA and anti-CD19 dual-target CAR-T cells (such as the BC19 construct developed by Xuzhou Medical University) are autologous chimeric antigen receptor (CAR) T cells engineered to simultaneously target B-cell maturation antigen (BCMA) and CD19. This dual-targeting approach is designed to overcome antigen escape and tumor heterogeneity, which are common mechanisms of resistance and relapse in single-target CAR-T therapies for plasma cell malignancies. BCMA is highly expressed on malignant plasma cells, while CD19 is expressed on B-cell progenitors and a subset of less-differentiated, stem-like myeloma cells. By targeting both, these CAR-T cells aim to achieve deeper and more durable responses. The therapy is primarily being evaluated in clinical trials for relapsed or refractory multiple myeloma and other plasma cell neoplasms, as well as autoimmune diseases like IgG4-related disease.
