anti-BCMA CAR-iNK is an experimental allogeneic cell therapy composed of natural killer (NK) cells derived from induced pluripotent stem cells (iPSCs) and engineered to express a chimeric antigen receptor (CAR) specific for B-cell maturation antigen (BCMA). Developed by Cell Inspire Biotechnology and partners, the therapy utilizes a non-viral piggybac transposon system to transduce iPSCs, which are then differentiated into highly pure NK cells (CD45+/CD56+). These CAR-iNK cells are designed to provide an off-the-shelf treatment for multiple myeloma, combining the targeted specificity of CAR-T therapies with the safety and scalability of iPSC-derived NK cells. Preclinical data presented at ASH 2023 demonstrated that these cells exhibit potent, dose-dependent cytotoxicity against BCMA-expressing myeloma cell lines such as NCI-H929 and OPM-2, although they were noted to be less efficient than GPRC5D-targeted counterparts in certain models.
