anti-BCMA CAR mRNA tLNP is an in vivo CAR-T cell therapy candidate developed by Aera Therapeutics for the treatment of plasma cell malignancies, such as multiple myeloma. The therapy utilizes a proprietary targeted lipid nanoparticle (tLNP) platform to deliver mRNA encoding a chimeric antigen receptor (CAR) specific for B-cell maturation antigen (BCMA) directly to CD8+ T cells within the patient's body. This approach aims to eliminate the need for ex vivo cell manipulation, expansion, or lymphodepleting conditioning, which are standard for traditional CAR-T therapies. Preclinical studies in humanized mice and non-human primates have demonstrated that the tLNP effectively delivers the CAR mRNA to resting T cells, resulting in dose-dependent tumor control and depletion of plasma cells in the peripheral blood and bone marrow.
