Anti-BCMA CAR-NK refers to a class of immunotherapeutic cell therapies that utilize natural killer (NK) cells genetically engineered with a chimeric antigen receptor (CAR) designed to target B-cell maturation antigen (BCMA). BCMA is a protein highly expressed on multiple myeloma (MM) cells, making it an attractive target for cancer therapy. These engineered NK cells are designed to recognize and kill BCMA-expressing cancer cells, often by secreting cytotoxic cytokines like IFN-γ and granzyme B. Different constructs may incorporate various intracellular signaling domains (e.g., CD28, 4-1BB, CD3ζ) and may ectopically express cytokines like IL-15 to enhance proliferation and persistence. This therapy is being developed as a potential treatment for multiple myeloma, offering advantages over CAR-T cell therapy such as a lower risk of cytokine release syndrome and the potential for "off-the-shelf" availability.
