anti-BCMA CAR-T (sdAb) is an autologous chimeric antigen receptor T-cell (CAR-T) therapy that utilizes a humanized single-domain antibody (sdAb) as its antigen-binding domain to target B-cell maturation antigen (BCMA). Unlike traditional CAR-Ts that use single-chain variable fragments (scFvs), the sdAb-based design aims for improved stability and potentially reduced immunogenicity. The construct incorporates 4-1BB and CD3ζ intracellular signaling domains to promote T-cell proliferation and cytotoxicity upon binding to BCMA-expressing plasma cells. Developed by Shenzhen Pregene Biopharma (often identified by the code PRG1801), this therapy has been investigated in investigator-initiated and Phase I clinical trials in China for the treatment of relapsed or refractory multiple myeloma (RRMM), demonstrating significant clinical activity including high rates of stringent complete response (sCR).
