Overview

Anti-BCMA CAR-T 9C4 is an academic, second-generation chimeric antigen receptor (CAR) T-cell therapy being developed as a low-cost adoptive cellular therapy candidate for multiple myeloma. The therapy utilizes a novel high-affinity single-chain variable fragment (scFv) derived from the 9C4 hybridoma clone, which targets the extracellular domain (ECD) of B-cell maturation antigen (BCMA). Molecular docking studies indicate that the 9C4 antibody binds to four novel epitopes (S, Q, N, E) on BCMA, interacting with 23 residues—significantly more than the 12-16 residues targeted by existing anti-BCMA antibodies. This increased binding complexity and high affinity are intended to improve therapeutic outcomes in patients with multiple myeloma, where BCMA is overexpressed and linked to poor prognosis. The project is led by researchers at the All India Institute of Medical Sciences (AIIMS), New Delhi.
