This investigational autologous dual-targeted chimeric antigen receptor (CAR) T-cell therapy is being developed by the Chinese PLA General Hospital for the treatment of relapsed or refractory IgG4-related disease (IgG4-RD). The therapy involves genetically modifying a patient's own T cells to express receptors that simultaneously target B-cell maturation antigen (BCMA) and CD19. This dual-targeting strategy is designed to comprehensively deplete pathogenic B-lineage populations, including CD19-positive B cells and plasmablasts, as well as BCMA-positive plasma cells that produce the pathogenic IgG4 antibodies characteristic of the disease. The therapy is currently being evaluated in a Phase 2 exploratory clinical trial (NCT07148791) for adult patients who have proven refractory to standard treatments such as glucocorticoids or rituximab.
