Overview

Anti-BCMA CD28z-4-1BBz CAR-iNKT is a third-generation chimeric antigen receptor (CAR) cell therapy that utilizes invariant natural killer T (iNKT) cells to target B-cell maturation antigen (BCMA). Developed by researchers at Imperial College London, this construct is engineered with a CAR containing two co-stimulatory domains—CD28 and 4-1BB—in addition to the CD3ζ activation domain. The use of iNKT cells as the cellular vehicle allows for an "off-the-shelf" allogeneic approach, as their invariant T-cell receptor does not cause graft-versus-host disease (GVHD). In preclinical studies for multiple myeloma, this specific third-generation construct demonstrated lower proliferative potential and reduced anti-tumor activity compared to second-generation CAR-iNKT variants, leading researchers to conclude that CD28z-based second-generation CARs may be more optimal for this platform.
