Overview

anti-BCMA CD28z CAR-iNKT is an allogeneic, "off-the-shelf" cellular immunotherapy being developed for the treatment of multiple myeloma. It consists of invariant Natural Killer T (iNKT) cells engineered to express a second-generation chimeric antigen receptor (CAR) targeting B-cell maturation antigen (BCMA), utilizing a CD28 costimulatory domain and a CD3ζ signaling domain. iNKT cells are a unique subset of T cells that are CD1d-restricted and do not cause allogeneic graft-versus-host disease (aGVHD), making them suitable for universal donor applications. Research conducted at Imperial College London suggests that the CD28z costimulatory configuration is optimal for the iNKT platform, providing superior effector-target avidity, immune synapse strength, and in vivo anti-tumor activity compared to 4-1BB-based or third-generation CAR designs.
