Anti-BCMA CD28z-OX40z CAR-iNKT is a third-generation chimeric antigen receptor (CAR) cell therapy engineered into invariant natural killer T (iNKT) cells. It targets B-cell maturation antigen (BCMA), which is highly expressed on malignant plasma cells in multiple myeloma. The CAR construct incorporates the CD28 and OX40 co-stimulatory domains alongside the CD3ζ signaling chain to enhance effector function, proliferation, and persistence. As an iNKT cell-based therapy, it utilizes the CD1d-restricted nature of iNKT cells to provide an off-the-shelf allogeneic product with a reduced risk of graft-versus-host disease (GvHD). This specific construct was evaluated by researchers at Imperial College London as part of a study to optimize CAR-iNKT platforms for the treatment of multiple myeloma, demonstrating high avidity and potent anti-myeloma activity in preclinical models.
