Anti-BCMA DAR-T is an investigational **allogeneic engineered T-cell therapy** developed by **Sorrento Therapeutics** for **multiple myeloma**. It uses donor-derived T cells genetically modified with a proprietary **non-viral vector knockout/knockin technology** to insert an **anti-BCMA dimeric antigen receptor** into the **TRAC** locus, thereby eliminating endogenous T-cell receptor expression and generating abTCR-negative cells while directing cytotoxic activity against **B-cell maturation antigen**-expressing malignant plasma cells. The lead construct reportedly incorporates a **third-generation signaling architecture** with **4-1BB, CD28, and CD3 zeta** signaling components, and preclinical studies showed potent in vitro and in vivo antitumor activity against BCMA-positive multiple myeloma models.
