Anti-BCMA OX40z CAR-iNKT is an investigational, preclinical cell therapy construct consisting of invariant natural killer T (iNKT) cells engineered with a second-generation chimeric antigen receptor (CAR) targeting B-cell maturation antigen (BCMA). This specific design utilizes the OX40 (CD134) co-stimulatory signaling domain fused to the CD3ζ activation domain. iNKT cells are a unique subset of T cells that express an invariant T-cell receptor (TCR) recognizing glycolipid antigens presented by CD1d; their lack of MHC-restricted TCR signaling makes them suitable for "off-the-shelf" allogeneic applications without the risk of graft-versus-host disease (GVHD). Developed primarily by researchers at Imperial College London, this therapy is being evaluated for the treatment of multiple myeloma, where BCMA is highly expressed on malignant plasma cells. Preclinical studies have compared this OX40z construct against other co-stimulatory designs (such as CD28z and 4-1BBz) to optimize effector-target avidity and immune synapse strength.
