Overview

Anti-BCMA universal CAR-T cells are an allogeneic chimeric antigen receptor (CAR) T-cell therapy targeting B-cell maturation antigen (BCMA), primarily developed for the treatment of relapsed or refractory multiple myeloma. Unlike autologous CAR-T therapies, universal (or allogeneic) CAR-T cells are manufactured from healthy donor T cells. These cells are typically genetically engineered to express a CAR specific for BCMA while simultaneously undergoing gene editing (e.g., using CRISPR/Cas9 or TALEN) to disrupt the T-cell receptor (TCR) alpha constant (TRAC) locus, thereby minimizing the risk of graft-versus-host disease (GvHD). This "off-the-shelf" approach allows for immediate treatment availability and standardized manufacturing, potentially overcoming the logistical and cost barriers associated with patient-specific cell therapies.
