Drug intelligence / Profile preview
Anti-C5aR1 antibodies are a class of monoclonal antibodies designed to target and inhibit complement component 5a receptor 1 (C5aR1, also known as CD88). C5aR1 is a G-protein coupled receptor primarily expressed on myeloid-lineage cells, including neutrophils, macrophages, and myeloid-derived suppressor cells (MDSCs). Activation of C5aR1 by its ligand, the potent anaphylatoxin C5a, drives chemotaxis, inflammatory cell recruitment, and the release of pro-inflammatory cytokines, contributing to the pathogenesis of various autoimmune and inflammatory diseases, such as ANCA-associated vasculitis (AAV), bullous pemphigoid, and giant cell arteritis (GCA), as well as tumor-promoting immunosuppression in oncology. By blocking the C5a-C5aR1 interaction, these antibodies inhibit downstream inflammatory cascades and neutrophil activation while sparing the potentially protective C5aR2 pathway. Several therapeutic candidates in this class, such as avdoralimab and izastobart, have been evaluated in clinical trials.
