Anti-CA IX antibodies are a class of monoclonal antibodies designed to target Carbonic Anhydrase IX (CA IX), a transmembrane protein that is highly overexpressed in various solid tumors, particularly under hypoxic conditions. CA IX plays a critical role in pH regulation, promoting an acidic extracellular environment that facilitates tumor invasion and survival. These antibodies are utilized across multiple clinical and research modalities: as diagnostic tools for immunohistochemistry (e.g., clone EP161), as imaging agents for PET/SPECT (often when radiolabeled with Zirconium-89), and as therapeutic vehicles for radioligand therapy (e.g., girentuximab labeled with Lutetium-177 or Actinium-225). In therapeutic contexts, they are primarily investigated for the treatment of clear cell renal cell carcinoma (ccRCC) and glioblastoma, where they can deliver cytotoxic radiation directly to tumor cells or inhibit the enzymatic activity of CA IX.
