Drug intelligence / Profile preview
Anti-cadherin-11 antibody refers to a class of monoclonal antibodies that target cadherin-11 (CDH11), a calcium-dependent cell-cell adhesion molecule primarily expressed on mesenchymal cells, such as fibroblasts, osteoblasts, and fibroblast-like synoviocytes (FLS). By binding to the extracellular domain of cadherin-11, these antibodies block homophilic cell-cell adhesion and disrupt downstream signaling pathways involved in tissue remodeling, inflammation, and metastasis. This therapeutic approach has been investigated for various indications, including rheumatoid arthritis, where cadherin-11 mediates joint destruction by FLS, and fibrotic diseases such as systemic sclerosis and pulmonary fibrosis. Additionally, anti-cadherin-11 antibodies have shown preclinical efficacy in preventing cancer metastasis, particularly prostate cancer bone metastasis, and in ameliorating chronic kidney disease and cardiovascular conditions like aortic valve stenosis. Notable clinical-stage candidates in this class have included RG6125 (formerly SDP051), which was developed by Adheron Therapeutics and Roche before being discontinued.
