The anti-caPCNA antibody is a diagnostic monoclonal antibody (or fragment, such as Fab2') that specifically targets the cancer-associated isoform of the Proliferating Cell Nuclear Antigen (caPCNA). While the standard PCNA protein is a central component of the DNA replication machinery expressed in all proliferating cells, the caPCNA isoform is uniquely expressed or post-translationally modified almost exclusively in malignant cells. This specificity allows the antibody to serve as a highly selective immunohistochemistry (IHC) biomarker for identifying various cancers, including breast, lung, colon, and ovarian carcinomas, across all clinical stages. Developed by researchers at the Indiana University Medical Center, the antibody is used to distinguish early-stage neoplasia from benign hyperplasia and is being investigated for its potential to predict patient outcomes based on expression levels in tumor tissues.
