Overview

An anti-CCL2 neutralizing antibody is a therapeutic agent designed to target and neutralize C-C motif chemokine ligand 2 (CCL2), also known as monocyte chemoattractant protein-1 (MCP-1). CCL2 is a key chemokine responsible for the recruitment of inflammatory monocytes and macrophages to sites of inflammation and tumor microenvironments. By binding to CCL2, the neutralizing antibody prevents its interaction with the cognate receptor CCR2, thereby blocking the recruitment of metastasis-associated macrophages (MAMs) and tumor-associated macrophages (TAMs). This pathway inhibition has been investigated for its potential to suppress tumor cell extravasation, metastatic seeding, angiogenesis, and chronic inflammatory or fibrotic conditions such as idiopathic pulmonary fibrosis and rheumatoid arthritis. Clinical-stage candidates developed under this mechanism include carlumab (CNTO 888) and ABN912.
