Anti-CCR2 monoclonal antibodies are a class of therapeutic agents designed to antagonize the C-C chemokine receptor type 2 (CCR2), a G protein-coupled receptor primarily expressed on monocytes, memory T cells, and dendritic cells. The primary ligand for CCR2 is CCL2 (also known as monocyte chemoattractant protein-1 or MCP-1). By blocking the CCR2-CCL2 axis, these antibodies inhibit the mobilization of inflammatory monocytes from the bone marrow and their subsequent recruitment into peripheral tissues or the tumor microenvironment. This mechanism is of therapeutic interest in treating chronic inflammatory diseases like rheumatoid arthritis, fibrotic conditions, and various malignancies where tumor-associated macrophages (TAMs) promote progression and immunosuppression. Notable examples in this class include plozalizumab (MLN1202/TAK-202), which has been evaluated in clinical trials for bone metastases and inflammatory disorders.
