Drug intelligence / Profile preview
Anti-CCR7 antibodies are monoclonal antibodies designed to target C-C chemokine receptor type 7 (CCR7, also known as CD197), a G-protein-coupled receptor that regulates the homing and trafficking of T cells, B cells, and dendritic cells to secondary lymphoid organs (SLOs) such as lymph nodes and spleen. CCR7 is highly expressed in various hematological malignancies, including chronic lymphocytic leukemia (CLL), mantle cell lymphoma (MCL), and other non-Hodgkin lymphomas (NHL), where it facilitates tumor cell migration into protective lymphoid niches that promote survival and drug resistance. By binding to CCR7, these antibodies block the interaction with its ligands, CCL19 and CCL21, thereby inhibiting chemotaxis, extravasation, and nodal homing. Additionally, some therapeutic anti-CCR7 antibodies, such as CAP-100, exert direct anti-tumor activity through Fc-mediated effector functions like antibody-dependent cellular cytotoxicity (ADCC) and complement-dependent cytotoxicity (CDC). Beyond oncology, targeting CCR7 is also being investigated for the prevention and treatment of graft-versus-host disease (GvHD) and autoimmune disorders by modulating the migration of alloreactive or autoreactive T cells.
