Anti-CCR9 CAR-T is an investigational chimeric antigen receptor (CAR) T-cell therapy designed to target C-C Motif Chemokine Receptor 9 (CCR9), a G-protein coupled receptor primarily expressed on T-cell acute lymphoblastic leukemia (T-ALL) cells. Unlike many other T-ALL targets, CCR9 has minimal expression on normal mature T and B cells, which reduces the risk of CAR-T fratricide and profound T-cell aplasia. The construct is a second-generation CAR utilizing a rat-derived anti-CCR9 single-chain variable fragment (scFv) linked to 4-1BB and CD3ζ signaling domains, delivered via a gamma-retroviral vector. Preclinical studies conducted by researchers at University College London, in collaboration with Autolus Therapeutics, have shown significant anti-leukemic activity and prolonged survival in patient-derived xenograft models.
