An anti-CD117 diabody is a recombinant, bivalent antibody fragment designed to target the Mast/stem cell growth factor receptor Kit (CD117), which is highly expressed on the surface of hematopoietic stem cells (HSCs). The diabody architecture consists of two single-chain Fv (scFv) fragments connected by a short peptide linker, which facilitates bivalent binding to CD117 while maintaining a significantly smaller molecular weight and shorter circulatory half-life compared to full-length monoclonal antibodies. This rapid clearance profile is specifically engineered for use in hematopoietic stem cell transplantation (HSCT) conditioning, where the goal is to transiently deplete host HSCs to create niche space for donor cells without the prolonged immunosuppression or systemic toxicity associated with traditional chemotherapy or radiation. Beyond its therapeutic potential in non-genotoxic conditioning, the anti-CD117 diabody is also utilized as a targeting scaffold for radiopharmaceuticals; for instance, when conjugated with Zirconium-89 ([89Zr]), it serves as a PET imaging agent to non-invasively map HSC niches and monitor c-Kit-positive malignancies.
