Anti-CD11b monoclonal antibodies are a class of biologic agents that target CD11b (Integrin alpha M / ITGAM), a subunit of the Mac-1 (CR3) integrin complex. CD11b is expressed on various immune cells, including monocytes, macrophages, granulocytes, and natural killer cells, where it mediates cell adhesion, migration, and immune signaling. In therapeutic development, these antibodies are explored for their potential to modulate the tumor microenvironment; for instance, neutralizing antibodies can inhibit the recruitment of pro-angiogenic bone marrow-derived cells following radiotherapy to prevent tumor recurrence. Conversely, agonist antibodies are being developed to promote the polarization of macrophages toward an anti-tumor M1 phenotype. While widely used as research reagents (e.g., clones ICRF44, 5C6), therapeutic applications are being investigated in oncology and autoimmune diseases such as rheumatoid arthritis.
