The anti-CD123 PGLALA antibody is a humanized monoclonal antibody targeting the interleukin-3 receptor alpha chain (CD123), which is overexpressed in various hematologic malignancies, including acute myeloid leukemia (AML) and blastic plasmacytoid dendritic cell neoplasm (BPDCN). This antibody is engineered with the 'PGLALA' triple mutation (L234A, L235A, and P329G) in the Fc region of the IgG1 scaffold. These specific amino acid substitutions are designed to silence Fc-mediated effector functions, such as antibody-dependent cellular cytotoxicity (ADCC), antibody-dependent cellular phagocytosis (ADCP), and complement-dependent cytotoxicity (CDC), by abolishing binding to Fcγ receptors and the C1q component of complement. Primarily developed and utilized by Janssen (Johnson & Johnson), this construct serves as a critical research tool and a control arm in the development of CD123-targeting bispecific antibodies, such as DuoBody molecules like JNJ-63709178. Its use allows for the evaluation of CD123 binding and receptor blockade independent of innate immune system activation, ensuring that therapeutic effects in bispecific formats are attributed to redirected T-cell activity.
