The **anti-CD146 nanobody** (specifically clone **112-2**) is a preclinical-stage single-domain antibody (nanobody) developed by researchers at **Shantou University** for the treatment of triple-negative breast cancer (TNBC). It specifically binds to the extracellular domain of melanoma cell adhesion molecule (MCAM/CD146) at leucine positions 331-332, inducing its internalization and proteolysis. By degrading CD146, the nanobody disrupts the crosstalk between adipocytes and TNBC cells in the tumor microenvironment, inhibiting IL-6-mediated JAK/STAT3 signaling and fatty acid oxidation (FAO). This dual action impairs lipid metabolic reprogramming and suppresses TNBC aggressiveness and lung metastasis *in vitro* and *in vivo*.
