Overview

anti-CD16-ovalbumin is an experimental antibody-antigen-conjugate (AAC) designed to target and reprogram neutrophils within the tumor microenvironment. The conjugate consists of a monoclonal antibody targeting the neutrophil-specific receptor FcγRIIIB (CD16b) linked to the model antigen ovalbumin (Ova). Upon intravenous administration, the AAC binds to neutrophils and induces their transformation into non-canonical antigen-presenting cells (nAPCs). These nAPCs effectively present the ovalbumin antigen to CD8+ T cells, bypassing dysfunctional dendritic cells and the tumor-draining lymph nodes by utilizing the spleen for T cell priming. This process triggers a robust anti-tumor immune response involving NK cells and memory T cells, leading to tumor regression and protection against metastases. It is being studied as a strategy to overcome immune exclusion and enhance the efficacy of checkpoint inhibitors like anti-PD-1.
