Overview

**Anti-CD19 + anti-BCMA CAR-T cells** are autologous T cells genetically engineered to express a bispecific chimeric antigen receptor (CAR) targeting both CD19 and B-cell maturation antigen (BCMA). This second-generation CAR therapy, often incorporating a 4-1BB costimulatory domain, enables T cells to recognize and eliminate malignant plasma cells expressing BCMA and a subset of less-differentiated myeloma cells expressing low levels of CD19, addressing antigen escape seen in single-target therapies. Primarily developed for relapsed/refractory multiple myeloma (R/R MM), clinical trials (e.g., Phase I/II) report high overall response rates (92%), complete response rates (62%), median progression-free survival of 19.7 months, and a favorable safety profile with manageable cytokine release syndrome.
