Overview

Anti-CD19 3rd generation CAR T cells is an investigational autologous cell therapy developed by Uppsala University for the treatment of relapsed or refractory CD19-positive B-cell malignancies, including B-cell lymphoma, B-cell leukemia, and Hodgkin lymphoma. The therapy consists of the patient's own T cells engineered via a GMP-grade retroviral vector (Moloney murine leukemia virus, MMLV) to express a third-generation chimeric antigen receptor (CAR) construct, SFG αCD19.28.4-1BB-ζ. This construct incorporates both CD28 and 4-1BB co-stimulatory signaling domains alongside the CD3-ζ activation domain, aiming to combine rapid tumor elimination with long-term T-cell persistence. The therapy has been evaluated in Phase I/IIa and Phase II clinical trials at Uppsala University Hospital in Sweden.
