Overview

anti-CD19-BB-ζ is a chimeric antigen receptor (CAR) construct designed to redirect the specificity of T lymphocytes towards tumor-associated antigens. It consists of a single-chain variable domain (scFv) derived from an anti-CD19 monoclonal antibody, which provides specificity for CD19-expressing cells. The construct also incorporates the hinge and transmembrane domains of CD8α, and the signaling domains of CD3ζ and 4-1BB. CD3ζ delivers the primary activation signal upon receptor engagement, while 4-1BB provides crucial co-stimulatory signals that enhance T-cell cytotoxicity and proliferation. This receptor is developed for the immunotherapy of B-cell non-Hodgkin's lymphoma (B-NHL), as B-NHL cells invariably express CD19. Preclinical studies have shown that T cells expressing anti-CD19-BB-ζ exhibit powerful cytotoxicity against CD19+ B-NHL cell lines and effectively suppress tumor growth in xenograft models, providing a rationale for clinical testing in patients with aggressive or relapsed B-NHLs refractory to conventional therapy.
