Overview

This investigational cell therapy consists of natural killer (NK) cells that have been genetically engineered to express a chimeric antigen receptor (CAR) targeting the CD19 antigen. Developed by St. Jude Children's Research Hospital, the CAR construct (anti-CD19-BB-zeta) incorporates an anti-CD19 single-chain variable fragment (scFv) linked to the 4-1BB (CD137) costimulatory domain and the CD3-zeta signaling domain. The NK cells, which can be autologous or allogeneic (haploidentical), are expanded ex vivo using a specialized manufacturing process involving a K562 myeloid leukemia feeder cell line genetically modified to express membrane-bound interleukin-15 (IL-15) and 4-1BB ligand. This expansion method is designed to enhance the proliferation, persistence, and cytotoxic activity of the NK cells. The therapy is primarily being evaluated in Phase 1 clinical trials for the treatment of relapsed or refractory B-lineage acute lymphoblastic leukemia (B-ALL).
