anti-CD19 CAR-imMac is a novel, off-the-shelf chimeric antigen receptor (CAR) macrophage cell therapy derived from induced pluripotent stem cells (iPSCs). It utilizes an immortalized precursor cell line (imMac) with a doxycycline-inducible system for c-MYC, BM1, and BCL-XL, enabling robust and scalable cell expansion. These imMac cells are transduced with a lentiviral vector encoding a CD19-specific CAR. Upon doxycycline removal, they undergo terminal differentiation into mature macrophages. The therapy is designed to overcome limitations of conventional CAR-T cell therapies, such as antigen escape, T-cell exhaustion, and manufacturing delays. It demonstrates enhanced phagocytosis and anti-tumor activity against CD19-overexpressing cancer cells, particularly when M1-polarized, and secretes inflammatory cytokines like TNF, IL-1β, and IL-6. Preclinical studies in xenograft models have shown significant inhibition of tumor growth and prolonged survival in B-cell lymphoma.
