Anti-CD19 CAR-NK cells are natural killer (NK) cells that have been genetically engineered to express a chimeric antigen receptor (CAR) targeting the CD19 antigen, which is commonly found on the surface of B-cell malignancies such as non-Hodgkin lymphoma and chronic lymphocytic leukemia. These therapies can be derived from allogeneic sources and may be further modified to express immunostimulatory cytokines like interleukin-15 for enhanced persistence and activity. Upon binding to CD19-expressing tumor cells, the CAR transmits an activation signal into the NK cell, leading to targeted cytotoxicity through mechanisms such as cytokine secretion and direct lysis of malignant B-cells. Anti-CD19 CAR-NK cell therapy is being developed as an off-the-shelf alternative to autologous CAR-T therapies with potentially lower risks of graft-versus-host disease or severe cytokine release syndrome[1][2][3][4][5].
