Overview

Anti-CD19 CAR-T cells (University of Utah) is an autologous chimeric antigen receptor (CAR) T-cell therapy developed for the treatment of relapsed or refractory CD19-positive B-cell malignancies, including non-Hodgkin lymphoma (NHL), acute lymphoblastic leukemia (ALL), and chronic lymphocytic leukemia (CLL). The therapy utilizes a second-generation CAR construct consisting of a murine anti-human CD19 single-chain variable fragment (scFv) derived from the FMC63 clone, linked to a 4-1BB (CD137) costimulatory domain and a CD3-zeta signaling domain. A notable feature of this program is the use of the Miltenyi Biotec CliniMACS Prodigy platform for automated, closed-system manufacturing (CellReGen process), which incorporates a transmembrane domain derived from TNFRSF19. This approach is designed to improve the consistency and scalability of CAR-T production within an academic clinical setting.
