Anti-CD19 CAR-T cells with immunosuppressive receptor knockdown is an autologous chimeric antigen receptor (CAR) T-cell therapy developed by The Sixth Affiliated Hospital of Sun Yat-sen University. The therapy consists of T cells engineered to express a CAR targeting the CD19 antigen, which is prevalent on B-cell malignancies. Additionally, these cells are modified using genetic knockdown techniques to reduce the expression of immunosuppressive receptors, aiming to enhance the persistence and anti-tumor efficacy of the CAR-T cells within the inhibitory tumor microenvironment. It is primarily being investigated in Phase 0/1 clinical trials for the treatment of relapsed or refractory B-cell malignancies, including diffuse large B-cell lymphoma (DLBCL) and follicular lymphoma.
