Overview

Anti-CD19 CAR-T cells with TNFAIP3 variants are an experimental cell therapy designed to treat B-cell malignancies. Developed by researchers at the Garvan Institute and Carina Biotech, these third-generation CAR-T cells are genetically engineered to express specific reduction-of-function variants of the *TNFAIP3* gene (also known as A20). TNFAIP3 normally acts as a negative regulator of NF-κB signaling; by introducing variants that reduce its activity, the CAR-T cells exhibit a lower activation threshold, increased secretion of effector cytokines, and enhanced cytotoxic activity. This approach aims to overcome T cell exhaustion and improve therapeutic persistence in the face of chronic antigen stimulation.
