Overview

Anti-CD19-SGRP CAR T cells are an experimental, engineered adoptive cell therapy designed to treat CD19-positive B-cell malignancies. These CAR T cells are "armored" to constitutively secrete a signal regulatory protein gamma-related protein (SGRP), a high-affinity SIRPγ-derived molecule that binds to CD47 on tumor cells. By blocking the CD47–SIRPα "don't eat me" checkpoint axis in a paracrine manner, the secreted SGRP promotes the phagocytosis of tumor cells by macrophages and microglia within the tumor microenvironment. This dual mechanism—direct T-cell mediated killing via the CD19 CAR and innate immune activation via CD47 blockade—aims to enhance anti-tumor efficacy and overcome resistance mechanisms like antigen escape or immunosuppression. The therapy has demonstrated superior efficacy in preclinical mouse models of CD19+ lymphoma compared to conventional anti-CD19 CAR T cells.
