anti-CD19-TCRζ-CD28 CAR-T cells are an investigational second-generation chimeric antigen receptor (CAR) T-cell therapy developed by The Second Affiliated Hospital of Henan University of Traditional Chinese Medicine. The therapy consists of autologous T cells engineered to express a CAR targeting the CD19 antigen, which is widely expressed on B-cell malignancies. This specific construct utilizes a CD28 costimulatory domain linked to the TCRζ (CD3 zeta) signaling chain to enhance T-cell activation, proliferation, and cytotoxic activity upon antigen binding. It is primarily being evaluated in the MatchCART clinical trial, a Phase I/II study that competitively infuses these cells alongside 4-1BB (CD137) costimulated CAR-T cells to compare their relative safety, efficacy, and engraftment potential in patients with refractory B-lineage leukemia and lymphoma, including acute lymphoblastic leukemia (ALL), chronic lymphocytic leukemia (CLL), and various non-Hodgkin lymphomas.
