Overview

anti-CD19-TCRz-CD137 CAR-T is an autologous, second-generation chimeric antigen receptor (CAR) T-cell therapy developed by The Second Affiliated Hospital of Henan University of Traditional Chinese Medicine. The CAR construct is composed of an anti-CD19 single-chain variable fragment (scFv) for tumor targeting, a CD137 (4-1BB) costimulatory domain to promote T-cell persistence and metabolic fitness, and a CD3-zeta (TCRz) signaling domain for T-cell activation. It is primarily investigated for the treatment of B-cell malignancies, including acute lymphoblastic leukemia (ALL), chronic lymphocytic leukemia (CLL), and various subtypes of non-Hodgkin lymphoma such as diffuse large B-cell lymphoma (DLBCL), follicular lymphoma (FL), and mantle cell lymphoma (MCL). In clinical research (notably the MatchCART study), this agent is often compared directly with CD28-costimulated CAR-T cells to evaluate differences in engraftment and long-term efficacy.
