Anti-CD20 CAR T-cell therapy is a type of adoptive cellular immunotherapy where a patient's T cells are genetically engineered to express a chimeric antigen receptor (CAR) targeting the CD20 protein (MS4A1), which is predominantly expressed on the surface of B cells. The CAR construct typically includes an anti-CD20 single-chain variable fragment (scFv) fused to intracellular costimulatory domains (such as 4-1BB or CD28) and a CD3ζ signaling domain. Upon infusion, these cells recognize and eliminate CD20-positive malignant or autoreactive B cells. This approach is primarily investigated for the treatment of relapsed or refractory B-cell non-Hodgkin lymphomas and chronic lymphocytic leukemia, and is being explored for autoimmune conditions.
