Overview

Anti-CD200 blocking antibodies are a class of immunotherapeutic agents designed to disrupt the interaction between the CD200 glycoprotein and its inhibitory receptor, CD200R. CD200 is an immune checkpoint molecule frequently upregulated in hematological malignancies, such as multiple myeloma (MM) and chronic lymphocytic leukemia (CLL), as well as some solid tumors. This interaction typically leads to the suppression of T-cell mediated immune responses and the induction of an immunosuppressive microenvironment. By blocking CD200, these antibodies prevent its binding to CD200R on immune cells, thereby reversing CD200-mediated immunosuppression and restoring the cytotoxic activity of T cells and other immune effectors against tumor cells. Research indicates that this blockade can reverse the phosphorylation of downstream signaling molecules like DOK2, which are involved in T-cell attenuation. Samalizumab (ALXN6000) is a notable clinical-stage example of this class.
