Drug intelligence / Profile preview
Anti-CD206 CAR T cells are an experimental, preclinical autologous chimeric antigen receptor (CAR) T-cell therapy designed to target and eliminate CD206-high (CD206+) profibrotic macrophages. Developed by researchers at Dartmouth College in collaboration with Celdara Medical, this second-generation CAR T-cell construct (anti-CD206-CD8TM-CD28-CD3zeta) utilizes a camelid-derived single-domain antibody (nanobody) targeting CD206 (macrophage mannose receptor, MRC1) fused to mouse CD28 hinge, transmembrane, and intracellular signaling domains, and a CD3zeta signaling domain. In preclinical models of systemic sclerosis (SSc), a single systemic dose of these CAR T cells successfully restored dermal white adipose tissue (DWAT) lost to fibrosis and reduced dermal thickness. Mechanistic studies suggest that CD206-high macrophages drive systemic sclerosis pathology by redirecting the lineage commitment of adipose-derived stem cells (ADSCs) from adipocytes toward myofibroblasts via an IL-6-dependent pathway.
