An anti-CD21 antibody is a monoclonal antibody designed to target and block Complement Receptor 2 (CD21), which is primarily expressed on B cells and follicular dendritic cells (FDCs). CD21 is a critical component of the B-cell co-receptor complex and plays a vital role in the activation and survival of B cells by binding to C3d-tagged antigens. In the context of autoimmune diseases like systemic lupus erythematosus (SLE), blocking CD21 can inhibit the expansion of self-reactive B cells and the formation of germinal centers, thereby reducing the production of pathogenic autoantibodies. Research suggests that dual blockade of CD21 on both B cells and FDCs provides the most potent suppression of autoantibody-secreting cells and ameliorates kidney pathology in lupus models.
