Overview

Anti-CD21 CAR-T is an experimental chimeric antigen receptor (CAR) T-cell therapy targeting CD21 (Complement receptor type 2), developed by Autolus and University College London for the treatment of T-cell acute lymphoblastic leukaemia (T-ALL). CD21 is expressed in a NOTCH-dependent manner in T-ALL and is primarily found on normal B cells, with minimal expression on mature T cells, which helps avoid the common issue of T-cell fratricide seen with other T-ALL targets like CD7 or CD5. The lead candidate utilizes a fragment antigen binding (Fab)-CAR architecture with a 4-1BB costimulatory domain, which demonstrated superior signaling and specificity compared to traditional scFv-based CARs. Preclinical studies in murine models have shown significant tumor control and survival advantages.
