Anti-CD226 monoclonal antibodies are a class of therapeutic agents designed to target CD226 (also known as DNAM-1 or PTA1), a co-stimulatory receptor expressed on natural killer (NK) cells, T cells, and platelets. CD226 plays a critical role in immune cell activation and adhesion by interacting with its ligands, CD155 (PVR) and CD112 (PVRL2). In the context of autoimmune diseases and organ transplantation, antagonistic (blocking) anti-CD226 antibodies are developed to suppress pathological immune responses by inhibiting effector T cell activation and promoting the expansion of regulatory T cells (Tregs). This mechanism is being investigated for conditions such as systemic lupus erythematosus (SLE), rheumatoid arthritis, and graft-versus-host disease (GvHD). Conversely, in oncology, the CD226 pathway is often targeted to enhance anti-tumor immunity, frequently in combination with TIGIT inhibitors. Several pharmaceutical companies, including Eli Lilly, Bristol Myers Squibb, and Novo Nordisk, have advanced specific anti-CD226 monoclonal antibodies into clinical development.
