Anti-CD25 depleting monoclonal antibodies are a class of immunotherapeutic agents designed to selectively eliminate CD25-expressing cells, primarily regulatory T cells (Tregs), within the tumor microenvironment. While early anti-CD25 antibodies like daclizumab and basiliximab were designed to block IL-2 signaling, newer "depleting" versions are engineered (often with enhanced Fc-gamma receptor binding) to trigger antibody-dependent cellular cytotoxicity (ADCC) or phagocytosis (ADCP). By depleting Tregs—which normally suppress the immune response—these antibodies aim to "release the brakes" on the immune system, allowing effector T cells to attack tumor cells more effectively. This approach is being investigated primarily in oncology, often in combination with PD-1/PD-L1 inhibitors, to overcome immune resistance in solid tumors.
