anti-CD25NIB is a non-IL-2-blocking, Treg-depleting monoclonal antibody targeting CD25 (the interleukin-2 receptor alpha chain, IL-2Rα). Unlike earlier anti-CD25 antibodies that block IL-2 signaling on both regulatory and effector T cells, anti-CD25NIB is engineered to selectively deplete regulatory T cells (Tregs) while preserving IL-2/STAT5 signaling in effector T cells. This selectivity enhances antitumor immune responses without the immunosuppressive side effects seen with previous CD25-targeted therapies. Preclinical studies have shown that anti-CD25NIB reduces tumor growth and increases effector T cell activation in vivo, especially when combined with other immunotherapies such as checkpoint inhibitors or cancer vaccines. The humanized version of this antibody (RG6292) has demonstrated efficient intratumoral Treg depletion and robust antitumor activity in both humanized mouse models and non-human primates, supporting its clinical development for cancer immunotherapy[6].
