Drug intelligence / Profile preview
anti-CD3 CAR is a preclinical chimeric antigen receptor (CAR) cell therapy being developed by iCell ImmunityX (formerly iCell Gene Therapeutics) for the treatment of aggressive T-cell malignancies, including peripheral T-cell lymphoma (PTCL) and T-cell acute lymphoblastic leukemia (T-ALL). The therapy utilizes a third-generation CAR construct containing an extracellular single-chain variable fragment (scFv) derived from the anti-CD3 monoclonal antibody OKT3, linked to CD28 and 4-1BB co-stimulatory domains and a CD3ζ signaling domain. Because CD3 is widely expressed on normal T cells, traditional CAR-T approaches face challenges with fratricide (self-targeting) and T-cell aplasia. To overcome this, the CAR construct has been engineered into natural killer (NK) cells (such as the NK-92 cell line) which do not express CD3, or into T cells with disrupted endogenous T-cell receptor (TCR)/CD3 expression. The CD3-directed CAR cells recognize CD3-positive malignant T cells and mediate their cytotoxic elimination.
