Drug intelligence / Profile preview
The anti-CD3 heavy chain-only antibody (anti-CD3 HCAb) is a preclinical-stage recombinant single-domain (VHH) antibody targeting the CD3ε subunit of the human T-cell receptor complex. Developed by Adimab, these humanized binders were discovered by combining llama immunization with a yeast-based HCAb presentation and discovery platform. They are designed as modular building blocks for T-cell-engaging (TCE) multispecific therapeutics, where the CD3-binding arm recruits T cells to eliminate tumor cells. Lacking a light chain, these HCAbs simplify the design and manufacturing of multispecific formats (such as bispecific antibodies or VHH-TCR fusions) while maintaining potent T-cell activation and tumor-killing efficacy with a favorable developability profile.
